Effects of selective vs. nonselective cyclooxygenase inhibition on dynamic renal potassium excretion: a randomized trial.

چکیده
Both selective and nonselective cyclooxygenase (COX) inhibitors can reduce potassium excretion and can produce or exacerbate hyperkalemia.1–12 We investigated whether there is a difference between the effects of nonselective COX-1/ COX-2 inhibitors and selective COX-2 inhibitors on provoked dynamic renal potassium excretion. We apply a mixed-effects model statistical approach that allows investigation of druginduced delays in reaching maximal potassium excretion, blunting/flattening of potassium handling curves, and shift/separation in potassium handling at peak potassium excretion.

ATP-dependent potassium channels are implicated in simvastatin pretreatment-induced inhibition of apoptotic cell death after renal ischemia/reperfusion injury
Background: Simvastatin is a widely used medication in cardiac care. Here we evaluate the role of ATP sensitive potassium (KATP) channels in simvastatin induced renal protection after renal ischemia/reperfusion (I/R) injury. Methods: A total of 81 male Wistar rats, were treated with simvastatin (10 and 20mg/kg/day gavage, one week). Some groups received glibenclami...
متن کاملSelective COX-2 Inhibitors: A Review of Their Structure-Activity Relationships
Non-steroidal anti-inflammatory drugs (NSAIDs) are the competitive inhibitors of cyclooxygenase (COX), the enzyme which mediates the bioconversion of arachidonic acid to inflammatory prostaglandins (PGs). Their use is associated with the side effects such as gastrointestinal and renal toxicity. The therapeutic anti-inflammatory action of NSAIDs is produced by the inhibition of COX-2, while the ...

متن کاملEffects of celecoxib and naproxen on renal function in the elderly.
OBJECTIVE To compare the effects of celecoxib, a cyclooxygenase 2-specific inhibitor, with the nonspecific cyclooxygenase 1 and 2 inhibitor naproxen on renal function in 29 healthy elderly subjects in a single-blind, randomized, crossover study. METHODS Subjects received either celecoxib, 200 mg twice daily, for 5 days followed by celecoxib, 400 mg twice daily, for the next 5 days, or they re...
متن کاملRole of cyclooxygenase-2 in the prolonged regulation of renal function.
The role of cyclooxygenase-2 (COX-2) in the prolonged regulation of renal function was evaluated during changes in sodium intake and reduction of NO synthesis. It was evaluated in conscious dogs by administering a selective inhibitor (nimesulide) during 8 consecutive days. Nimesulide administration to dogs with normal or high sodium load did not modify glomerular filtration rate but reduced ren...
